Abstract
The time course of incorporation of [8-14C]adenine into purine bases of different RNA fractions in growing HeLa S3 cells has been studied. A high-molecular-weight RNA fraction, which resembles closely the microsomal RNA in HeLa cells, appears to be the end product in the biosynthesis of RNA. The data strongly suggest the nuclear origin of microsomal RNA in growing HeLa cells, with an RNA fraction (nuclear RNA) present in the nucleus as a polynucleotide precursor. A simple kinetic model is proposed, which under certain restrictive conditions may be used to define the "intracellular transport rate" of RNA from nucleus to cytoplasm.
