Abstract

B-ALL patients with intrachromosomal amplification of chromosome 21 (iAMP21) have an inferior outcome with standard therapy, which improves markedly, when treated as high risk disease using intensive chemotherapy. The identification of three or more extra copies of RUNX1 gene on chromosome 21, by FISH, is used to define iAMP21. Multiplex ligation-dependent probe amplification (MLPA) is emerging as a high throughput screening assay for iAMP21 and may act as alternative to comparative genomic hybridization.
