Abstract
There is evidence for the presence of ntrC and nifA like genes and their involvement in the expression and regulation of nitrogen fixation (nif) genes in Azospirillum. This is supported by the isolation of our group of Tn5 induced mutant of A.brasilense which exhibited NifA phenotype. Tn5 mutagenized nif DNA of this mutant (Nif 27) was, earlier, cloned in the recombinant plasmid pMS188. A deletion derivative of pMS188 has been created to remove most of the Tn5 DNA present on a HpaI fragment. The wild type nif regulatory gene seems to be located on a 12kb EcoRI fragment. Homology of this nif regulatory region has been detected in several strains of A.brasilense and A.lipoferum.
