Abstract

A series of 1,2,3,4-tetrahydroisoquinoline substituted 2H-pyranones and 1,3-teraryls have been synthesized taking berberine as lead and evaluated for glucose uptake stimulatory effect in skeletal muscle cells. Compound 5d showed significant stimulation of glucose uptake in a concentration-dependent manner with maximum stimulation (2.7 fold) at 10 µM in L6 skeletal muscle cells. The compound was found to have synergistic effect with insulin, and mechanistic studies revealed the PI-3-K-mediated signaling pathway for stimulation of glucose uptake in skeletal muscle cells.
