Abstract

Protoplast fusion was achieved between the cells of like mating type, in a citric acid-producing strain of Yarrowia (= Saccharomycopsis) lipolytica. Protoplasts were derived from auxotrophic mutants obtained from this strain, carrying identical mating type. Upon regeneration, the fusion products gave rise to prototrophic diploids. Analysis of 57 meiotic segregants of one diploid, carried out in detail, revealed 79% of recombinant phenotypes. Further study of citric acid production among these segregants confirmed that genetic recombination can be achieved by the fusion of protoplasts of identical mating type in this industrially important strain.
