Abstract

The production of monohapoid Solanum tubersum embryoids and plantlets with 2n=12 chromosomes from microspores cultured within anthers, is reported. By selecting the precise developmental stage of microspores and by modifying the culture media, it was possible to achieve microspore embryogenesis in 35% of the plated anthers. Potato extract was found to be beneficial, but the response could be obtained even in Murashige and Skoog [1] medium. For maximum response, presence of 6% sucrose, 0.5% activated charcoal and 4×10−6 M 6-benzylaminopurine (BAP) was necessary. The cytological examination of young embryoids and of root tips of developed plants revealed monohaploids as well as dihaploids.
