Abstract

A fluorometric method has been described that allows the microdetermination of glycolytic intermediates in extracts from tissues or celis. The method utilizes commercially available crystalline enzymes of glycolysis coupled finally to a pyridine nucleotide-linked dehydrogenase so that the time course of the reaction is automatically recorded. Illustrations have been presented showing the applicability of the method to the quantitative assay of intermediates down to a fraction of a micromolar range of concentration in the assay system.
