Abstract

The present study describes the application of a commercial fluorimeter to the enzymic assay of pyridine and adenine nucleotides in neutralized extracts of cellular material. The method retains the advantage of recording the reaction kinetics during the assay with the increased sensitivity associated with the fluorimetric method. The routine application of the method to the detection of 0.1 to 1.0 µmolar concentrations of nucleotides in extracts of yeast cells, liver mitochondria, and submitochondrial particles has been illustrated. In addition, various facets of the assay procedure have been discussed as well as problems associated with the determination of optimal conditions for the application of protein precipitants to terminate the reaction.
