Abstract
An improved method to synthesize enamide-containing macrolides by copper-mediated intramolecular vinylation of amides is described. The method was applied to access several large-ring macrolides, including neuroactive natural product palmyrolide A, desmethyl palmyrolide A, and homologated sanctolide A.
